Learn How to Culture (Isochrysis galbana). #OJT

Thursday, August 30th 2018. Still about mussel, we were started our OJT today with siphoned all the nursery tanks, around 8 tanks. We were guided by our mentor, and also learned how to siphon the mussel tank based on protocol.




After that we lunch, then counted the algae again which is Isochrysis galbana, by using hemocytometer and here are the results :
  • Isochrysis galbana

A block : 1243 cells
B block : 1445 cells
C block : 1494 cells
D block : 857 cells

Corner Block=(1243+1445+1494+857)/4 〖 × 10〗^4
= 12.597.500 cells/ml

Today we feel more harder to count the algae, because of size of the algae and the amount of the algae, but its ok, we could manage.
After counted the algae we were tried to culture the algae (Isochrysis galbana) this phytoplankton is the new species that we learned from this OJT. We don’t have something like this in our campus because we live in the middle of the island and far from the sea, and Isochrysis galbana, Chaetoceros sp., and Tetraselmis sp., are kind of sea water’s phytoplankton. 
Procedures of culture Isochrysis galbana :
  • The tool that will be used need to disinfected first.
  • Add 250 ml sea water into the long neck.
  • Add 200 ml of the culture of phytoplankton into the bottle (starter).
  • Add 250 ml sea water again.
  • Add 1 ml of fertilizer (CONWY)
  • After that add the air hose for the aeration and cover it with cotton as a filter from any dirth things, and then cover it again using aluminum foil for more savety and also used an air connector for connecting each air hose, than we connect air hose to the aeration. Don’t forget to use a label, and we can scale up after 3 days culture.

After finished our OJT we come back, dinner, and rest.
