Scale Up the Algae #OJT

Wednesday, August 24th 2018. Remember my first day of OJT?. Actually at that day we are cultured Chaetocerus sp. And Tetraselmis sp. So today is our scale up day! from the three bottles that we cultivate it become six bottles (actually nine bottles but we don’t use the othere one). Here the picture.


Than we’ll scale up our second culture on tuersday aka three days later. After the scale up was finished than we are looked around the hatchery to visit our OJT’s place for next week. After that we lunch and dzuhur prayer.
then we take 1 ml of the sample from Chaetocerus sp. And Tetraselmis sp., than we count the results, than we get this :
we use Hemocytometer for count the brown algae (Chaetocerus sp. ) and the green algae (Tetraselmis sp.). and with this formula

  • The brown algae (Chaetocerus sp.) : 

A block : 236 cells
B block : 224 cells
C block : 204 cells
D block : 244 cells

Corner Block=(236+224+204+244)/4 〖 × 10〗^4
= 2.270.000 cells/ml
  • The green algae (Tetraselmis sp.) :

A block : 29 cells
B block : 51 cells
C block : 56 cells
D block : 43 cells
Corner Block=(29+51+56+43)/4 〖 × 10〗^4
= 447.000 cells/ml



It make sanse because the green alga more bigger than the brown alga, and brown algae is used to feed shrimp in nauplii stage, and the green algae is used to feed shrimp in zoea until mysis or post larvae stage.
After we count our algae than we discuss about yesterday’s OJT with my partner than we go back to our boarding school by using tri cycle.
